Molecular mechanisms of Bacillus subtilis in response to cadmium stress under biochar addition |

Abstract: |
To explore the molecular mechanism of Bacillus subtilis in response to cadmium stress before and after biochar addition, based on transcriptome sequencing, this study compared and analyzed the differences in gene expression of B. subtilis in four systems(normal, cadmium, biochar, and biochar-cadmium system). The results revealed that there were 1 932 differentially expressed genes in B. subtilis under cadmium stress, including 967 up-regulated genes and 965 down-regulated genes. The metabolic activity of B. subtilis was significantly inhibited. However, its flagellum assembly and bacterial chemotaxis pathway were significantly enhanced. The gene expressions of ion transport and cationic transmembrane transporter functional items were up-regulated, suggesting that B. subtilis avoids cadmium toxic pressure by up-regulating the expression of genes related to cell movement and ion transport. Compared with the cadmium system, 1 541 differentially expressed genes were produced in the biochar-cadmium system, including 691 up-regulated and 850 downregulated genes. The presence of biochar enhanced the expression of bioprocess, cell components, and molecular function genes in B. subtilis. The gene expressions related to substance and energy metabolism in B. subtilis were up-regulated. The expression of the cadmium resistance gene czcD and cadA and the membrane function-related differential gene in B. subtilis was mainly regulated. These results indicated that biochar could alleviate the toxic effect of cadmium stress on B. subtilis and stimulate the metal resistance of B. subtilis. |
